"Niacin metabolism does consume methyl donors (SAMe), however, the magnitude of the "sink" effect is dose dependent. Niacin metabolites are excreted as methylated pyridones produced from a methylated intermediate called N-methylnicotinamide. Basu and Mann showed that a homocysteine levels increased with pharmacological doses of niacin in a rat model. The authors investigated the methionine cycle effects associated with high does niacin use for lipid management. The methyl donor depletion effect on homocysteine was dose dependent, as the control group (20 fold lower dose) showed virtually no change in homocysteine levels. As a general rule of thumb as long as you are using 50 mg or less per day of combined niacin for adult dosing and there are sufficient methyl donors being supplied (which you do) then it is unlikely that you are going to deplete the methyl pool to an appreciable extent. Clinically, I begin to see homocysteine levels increase when patients are self treating at >500 mg per day of niacin to support lipid metabolism."
